Validated processes and automation for standard-compliant production

  • Pre-use post sterilization integrity testing (PUPSIT) is mandatory for product filters according to Annex 1 (§ 8.82 ff.).
  • Maximum pore size of product filters: 0.2 or 0.22 μm
  • The test must be performed in-place (with the filter installed in the system).
  • Operators are guided via a shadow board that indicates the correct positions for each component of the single-use set.
  • Fully-automated testing eliminates manual testing and minimizes sources of potential errors.

Validation and Process Reliability in Freeze-Drying

Reliable sterilization of freeze dryers – validated processes for maximum purity

Annex 1 emphasizes the need for scientifically sound validation of sterilization processes. Sterilization cycles, for example, require precise monitoring of temperature, pressure, and time, as well as effective air removal to ensure complete sterilization. Routine operating parameters must be strictly maintained and thoroughly documented.

 

Freeze dryer sterilization at OPTIMA

  • Equipment and aseptic piping are sterilized before each freeze-drying cycle using clean steam in accordance with EN 556-1.
  • Complete process control is ensured through temperature monitoring and precise pressure regulation, including monitoring of process sequence and sterilization times:
  • Monitoring of sterilization temperature
  • Temperature measurement at defined points along the chamber/condenser outlet, filter test units, and pumps.

Precise Leak and Process Monitoring

According to Annex 1, systems such as hydraulic units, heating, and cooling systems should, where possible, be located outside the processing area. Any leaks that may pose a risk to the product must be detectable via an appropriate indication system.

Custom-Tailored, Fully Automated Loading and Unloading Systems for Freeze Dryers

According to Annex 1, robotics and automation should also be used in freeze-drying processes to avoid critical human intervention – for example, through automated loading and unloading of freeze dryers. The transfer of partially closed containers must always occur under Grade A cleanroom conditions and with minimal operator involvement.
